    For the determination of vitamin B_6 derivatives, at least, 6 biologically active vitamers (PL, PN, PM and these phosphate esters) together with pyridoxic acid (PIC), an HPLC system composed of a single reversed-phase ODS column and a highly sensitive fluorescence detector by an isocratic solvent was proposed. Relatively low fluorescence intensity of intact PLP under acidic conditions was improved by converting it to pyridoxic acid 5'-phosphate (PIC-P) in the addition of 5mM KCN at alkaline pH and subsequent heat treatment at 50℃ for 3h. By using the method, the vitamin B_6 contents in some foods and biologically interested materials were analyzed and these data were compared with the data obtained from microbial assay using Saccharomyces cerevisiae ATCC 9080.

    The serum vitamin B_6 (PAL, PAM, PIN) and PLP levels in 54 pregnant women and in 43 controls of non pregnant young women were investigated by HPLC method. The mean serum B_6 levels in pregnant women showed 6.7±4.3ng/ml, while that of controls showed 12.0±3.2ng/ml (p<0.001). The mean serum PLP levels in pregnant women showed 3.9±1.9ng/ml, while that of controls showed 9.3±3.7ng/ml (p<0.001). The decrease of serum vitamin B_6 and PLP levels occurred gradually during pregnancy, and lowest levels were shown during the last trimester. The dietary B_6 intake in 75 pregnant women estimated by evaluating three-days records was 2.1±0.7mg/day. The serum B_6 levels in 13 women estrgen-containing oral contraceptive was 7.7±3.8ng/ml (p<0.001). This suggests that estrogen play a role in decrease of serum B_6 levels during pregnancy.

    Niacin metabolism in omnivora, carnivora and herbivora was compared with high-performance analysis of urine. In omnivora (ten kinds of primates and pig), excreted amount of N^1-methyl-2-pyridone-5-carboxamide (2-Pyr) occupied 60-90% of the total urinary excretion of NiA-NH_2 and its metabolites. So, it was found that NiA-NH_2 was catabolized as follows in omnivora; NiA-NH_2→N^1-methylni-cotinamide (MNA)→2-Pyr. In carnivora (cat, dog, lion and tiger), the urinary excretion of MNA occupied above 90% of the NiA-NH_2 and its metabolites totally excreted and the oxidized forms of MNA such as 2-Pyr and N^1-methyl-4-pyridone-3-carboxamide were very slight. So, NiA-NH_2 was found to be catabolized as follows in carnivora; NiA-NH_2→MNA. In herbivora (four kinds of artiodactyla and horse), the urinary excretion of NiA-NH_2 occupied 80-100% of the total urinary excretion of NiA-NH_2 and its metabolites. Thus, it was found that NiA-NH_2 was hard to be catabolyzed in herbivora. In all mammals examined here, the urinary excretion of NiA, NuA and NiA-NH_2 N-oxide was below the limit of detection.

    Prototype of nicotinamide-biosensor system was constructed, for the first time, with partially purified nicotinamidase (EC 3.5.1.19) from "Toramakitake" mushroom, ion sensitive field effect transistors (ISFET), a differential amplifier, a high sensitive potentiometer and a personal computer. The enzyme ISFET and reference ISFET were prepared by immobilizing the enzyme and bovine serum albumin on each ISFET. The ultra small change of pH on the enzyme ISFET, which was caused by nicotinamidase reaction dependent on the nicotinamide concentration, was measured potentiometrically. The differential voltage (mV) between both ISFET at stationary phase was plotted against the logarithm of nicotinamide concentration (molarity) to obtain calibration curve. As the result, linear relationship was observed among 60-160mM of nicotinamide with a relation coefficient of 0.989. Possibility for practical use of this new type of nicotinamide sensor was suggested.
