The effects of ascorbic acid (AsA) and copper on the activity of NADPH-cytochrome c reductase in the liver microsome of guinea pigs were investigated. The enzyme activity was depressed by depletion of AsA. The administration of 500 ppm copper in the drinking water also inhibited the enzyme activity and decreased the amount of AsA in the liver. In the in vitro experiment, 3.8×10^<-6>M of copper inhibited the enzyme activity. The results of these experiments showed that inhibition of the enzyme activity by copper was caused by the direct inhibitory effect of copper ion and also due to the decrease of AsA content.

In previous paper, synthesis of vitamin D_2 tetraacetylglucoside was reported. The present work was concerned with the stability of this derivative under various storage conditions. Crystalline vitamin D_2 tetraacetylglucoside (D_2 TAG) were sealed in glass ampules in the presence of either air or nitrogen gas, and then stored for 175 days in a freezer (-20℃), incubator (40℃), or a room under either dark or light (avoid direct sunlight). Observations on coloring of the crystals, determinations of melting points, developments by thin-layer chromatography, measurements of UV-and IR-absorption spectra and column chromatographic analyses were carried through the storage period. Vitamin D_1 stored under the identical condition was examined as controls. From these results, it was found that vitamin D_2 TAG was more stable than vitamin D_2.

The human plasma 25-Hydroxyvitamin D_3 (25-OH-D_3) was successfully measured by the competitive protein binding assay using the serum or kidney cytosol derived from the vitamin D deficient rat as a binding protein. The standard 25-OH-D_3 could be assayed proportionally from 0.5 to 5.0 ng. The mean plasma 25-OH-D_3 concentration of healthy infants and children was 21.6±10.1 ng/ml and that of mature newborn infants within 48 hours after delivery was 3.5±1.9 ng/ml.

In order to study on the conversion of provitamin D into vitamin D in vitro by sunlight, a procedure for irradiating an ergosterol (a provitamin D) solution and determining vitamin D_2 in the irradiated solution was proposed. Twenty-five ml of an ergosterol solution in ethanol (0.5 mg/ml) containing di-tert-butylhydroxytoluene as an antioxidant (0.1 mg/ml) was exactly taken in a quartz-made erlenmeyer flask and allowed to stand at an outdoor place between 11 and 14 o'clock for the sake of irradiating by sunlight. The yield of vitamin D_2 in the irradiated solution was determined according to the method applying the gas-liquid chromatographic procedure as reported previously. This method was confirmed to be an excellent one because of showing accuracy and good reproducibility even though yield was about 2%, and therefore it must be available for the author's purpose.
