    α^4-Norpyridoxine 5'-phosphate (4-NOP-P) was synthesized and its antivitamin B_6 activity in vitamin B_6 apo-enzyme systems was tested. 4-NOP-P as well as 4-deoxypyridoxine 5'-phosphate (4-DOP-P), showed a competitive inhibition against pyridoxal 5'-phosphate in the reaction systems using aspartate aminotransferase from baker's yeast and tyrosine aminotransferase from rat liver (K_i=0.45×10^<-7>M and 0.7×10^<-7>M, respectively). 4-NOP-P exerted a noncompetitive inhibition (K_i=3.7×10^<-6>M) on tryptophanase from Escherichia coli, while 4-DOP-P showed no inhibition. α^4-Norpyridoxine exerted no effect on pyridoxine dehydrogenase from baker's yeast.

    O, S-Dibenzoyl thiamine hydrochloride (DBT-HCl) was tested by the rec-assay and the reversion assay. In the rec-assay with Bacillus subtilis strains H17 (Rec^+) and M45 (Rec^-), DBT-HCl gave negative results at a dose of 10mg/disk. When Escherichia coli strains B/rWP-2 and B/rWP-2hcr^-, and Salmonella typhimurium strains TA1535,TA1537,TA98 and TA100 were used in the reversion assay, DBT-HCl was not mutagenic in any of the indicator strains at concentrations ranging from 500 to 5,000μg/plate, irrespective of the treatment with mammalian metabolic activation system (S-9 mixture) prepared from phenobarbital-treated rat. Negative results were also obtained for sodium benzoate and sodium hippurate, the major metabolite of DBT, in the same test systems as above. From these results, it is concluded that DBT-HCl is not mutagenic under the present experimental conditions.

    O, S-Dibenzoyl thiamine hydrochloride (DBT-HCl) was examined for the mutagenicity by the dominant lethal test and in vivo cytogenetic test. In the dominant lethal test, (C3H × SWV/V) F_1 male mice were orally given a single dose of 1,000mg/kg or five successive doses of 300mg/kg/day of DBT-HCl. Mean numbers of viable implants per female in the experimental groups did not differ significantly from those in the vehicle (5% gum arabic solution) at any periods examined. In vivo cytogenetic effects of DBT-HCl were examined on bone marrow of CF#1 male mice receiving the similar treatment as in the dominant lethal test. There was no increase of chromosome aberration in any group treated with DBT-HCl as compared with the contro1. These results indicate lack of mutagenicity of DBT-HCl in the present experimental conditions.

    Intrinsic factor (IF) in rat gastric mucosal extract was bound efficiently to vitamin B_12-Sepharose without significant changes in its nature to produce IF-vitamin B_12-Sepharose. Purification of intestinal receptor for IF-vitamin B_12 complex was performed by affinity chromatography using the IF-vitamin B_12-Sepharose as an affinity adsorbent with recovery of 48.5% and specific activity increased 335 folds of original sample.

    The development of rachitis was observed on albino rats of Wistar strain which were resistant to vitamin D depletion. The development of rachitis was assessed from the width of the epiphyseal plate in radius and serum inorganic phosphorus level. Three weeks depletion period, which has been recommened for the line test, did not show clear evidence of rachitis in Wistar strain rats. Six weeks depletion period was shown to be suitable, namely, the width of the epiphyseal plate was widen to 1.50 mm and serum inorganic phosphorus level was reduced to 3.4 mg/dl. Therefore, the effects of vitamin D_3 and 1α-hydroxyvitamin D_3 (1α-OH- D_3) on the healing of rachitis were observed on Wistar strain rats which were raised on rachitogenic diet for 6 weeks. In good agreement with the findings of other investigators which Holzman strain rats were raised on rachitogenic diet for three weeks. 1α-OH-D_3 was more effective than vitamin D_3 for the healing of rachitis and the elevation of serum inorganic phosphorus level.
