    Many reports on the relationship between tryptophan (Trp) and niacin (NiA) have been published since Elvehjem and Goldsmith. Also the relationship between leucine (Leu) and NiA was studied by Gopalan et al. Goldsmith and Horwitt reported that 60 mg of Trp is equivalent to 1 mg of NiA. However, this ratio are not stationary. The relationship between Trp and NiA is affected when the physical conditions, the amount of Trp intake, caloric intake, and certain amino acid imbalances are varied. Gopalan et al. reported that excess intake of Leu induced the same biochemical effect in the human and other mammals as in pellagrin, and amino acid imbalance due to an excess of Leu has been suggested as a possible factor in the development of pellagra. On the other hand, Truswell et al. did not support the suggestion that a relative excess of Leu in the diet played a part in the pathogenesis of pellagra. In our experiments, there was not seen the same biochemical effect as in those of Gopalan et al., but a marked decrease of the plasma valine level was observed, when supplied large amounts of Leu.

    In order to study the interaction between B_<12> derivatives and SH groups of protein, the effect of B_<12> derivatives on the activity of thiol enzymes, papain and yeast alcohol dehydrogenase (YADH), was investigated. Both papain (7×10^<-2>mM) and YADH (1.4×10^<-3>mM) were inhibited by the incubation with 5.0×10^<-2>mM to 1.0×10^<-2>mM of aquocobalamin (OH-B_<12>) or cyanocobinamide. The inhibition of these thiol enzymes with OH-B_<12> was increased rapidly at pH greater than 6.0. Under anaerobic conditions, papain was scarecely inhibited, while YADH was inhibited as much as under aerobic conditions. The inhibitory effect of OH-B_<12> on YADH was merkedly decreased by the addition of NAD or NADH in the reaction medium. The inhibited papain was reactivated by KCN treatment. On the other hand, the inhibited YADH was not significantly reactivated by cysteine. The SH groups of the inhibited enzymes were found to decrease in comparison with those of the native enzymes.

    A simplified method was established for assay of intrinsic factor in rat gastric juice by modification of the zirconium phosphate gel technique of Hansen et al. Examination of various assay conditions, especially adjustment of the pH at the adsorption process to 5.0,provided many advantages : better preservability of zirconium gel once prepared ; clear-cut distinction between active intrinsic factor and inactive one ; good reproducibility of assay values ; availability of a small amount of gastric juice ; and a relatively simplified technique.

    The reaction of the thiazolium ylide carbanion at 2-position in thiamine with various diethyl acylphosphonates such as diethyl alkionyl, aroyl, aralkionyl and thenoyl phosphonates, afforded thiazinochrome derivatives (IIIa-h, IVa-h) in good yields respectively, which were hydrolysed to give corresponding thiazinothiamine derivatives (Va-h, VIa-h). The UV and NMR spectra about III-VII were also discussed.

    It is well known that thiaminase I catalizes the transfer of the pyrimidinyl group from various onium compounds containing (4-amino-2-methyl-5-pyrimidinyl) methyl group to 4-methyl-5-(β-hydroxyethyl) thiazole (Th) to form thiamine. Some properties of the thiamine synthesis with N-pyrimidinyl pyridoxinium-3-phenolate (Pm-PIN) and Th by the purified thiaminase I from Cl. sporogenes ATCC 8075 were investigated. The optimal pH of the reaction was 6.0 and the optimal temperature was 20℃. The enzyme was completely destroyed by heating for 10 min at 70℃. Thiamine synthesizing ability of the enzyme was accelerated with sulfhydryl compounds and inhibited with SH reagents, as in the case of the thiamine decomposing reaction. The inhibition by p-chloromercuribenzoate was completely restored by dithiothreitol. The effects of metal ions on the thiamine synthesis was the same as that on the thiamine decomposition. The K_m values in the thiamine synthesis at 20℃ (pH 6.0) in the presence of dithiothreitol were 3.3×10^<-5>M for Pm-PIN, 3.0×10^<-5>M for N-pyrimidinyl pyridinium bromide hydrobromide, and 1.11×10^<-4>M for Th.
