We have found by using gel filtration and [^<14>C]riboflavin that ester forms of riboflavin are synthesized fairly actively from riboflavin in the rat lens. Riboflavin was incorporated into single whole lens by simple diffusion and then converted to the ester forms with subsequent partial binding to lens protein. Change in the amount of high molecular weight phosphate compounds, especially RNA, was found in B_2 deficient rat lens. Lenticular riboflavin-binding capacity and the percentage of high molecular weight protein aggregates were increased remarkably at pH 9.0 as compared to those at pH 4.0 and 7.0. They were further promoted by exposure to light in the presence of riboflavin as compared to controls incubated in the dark. It has been well established that the lens is one of the organs in which riboflavin is metabolized and that lenticular riboflavin has two functions; coenzyme and photosensitivity.
