The pharmacological effects of FAD on the heart, peripheral blood vessels, arterial pressure, and intestinal canal were observed on rabbits and the results were compared with those of adenylic acid and FMN. FAD has a marked depressive action on the blood pressure, an inhibitory action the movements of the intestinal canal, a constrictive action on the ear blood vessels, and an inhibitory action on the heart beat of the rabbit. These pharmacological actions of FAD are mainly ascribable to its adenylic acid component, and through the combination with FMN these actions are strengthened and prolonged.

An acid clay dried at 105-110℃ was packed in a chromatographic column, 6 cm in height and 0.7 cm in diameter. At first 15 ml of 7% ethanol-hexane was passed through the colomn, followed by 10 ml of 10% ethyl ether-hexane. The column thus treated could be used to separate vitamin D_2 from vitamin A. The recoveries of vitamin D_2 were 93-94%. The amount of vitamin A in the sample could not exceed 800 I. U.. A small amount of destruction products of vitamin A was found to be eluted in the vitamin D_2 fraction, thus samples with less than 400 I. U. of vitamin D_2 could not be applied to this chromatography. Cholesterol could be separated from vitamin D_2 by the above procedure.

Eight kinds of commerical acid clays were tested. They were dried at 105-110℃, packed in a chromatographic column, 6 cm in height and 0.7 cm in diameter, and washed preliminarily with 15 ml of 7% ethanol-hexane (using 96% ethanol) followed by 10 ml of 10% ethyl ether-hexane. Four out of the eight clays gave good results. In the chromatography using one of these activated acid clays, four kinds of developing solvent systems (n-hexane, carbon tetrachloride, benzene, 10% ethyl ether) were tested. The recoveries of added vitamin D_2 were better when n-hexane and 10% ethyl ether-hexane were used then carbon tetrachloride and benzene were used. When n-hexane alone was used as a developing solvent, vitamin D_2 was present in the 4-50 ml of the effluents. The amount of vitamin A which could be introduced onto the column was found not to exceed 7,000-8,000 I.U.

Comparisons were made in the producing abilities of riboflavin (FR), flavinmononucleotide (FMN) and flavinadeninedinucleotide (FAD) of about a hundred strains of Aspergillus flavus-oryzae. No FMN was found in culture broth and no FR was observed in mycelium. Furthermore, the tendency was observed that the strains of such kind as mycelium of slow growth, conidia of unsatisfactory formation and short conidiophores, produced rather large amounts of flavin-substances. A strain producing the highest level of FAD, 49μg per 1g of dry mycelium, was selected from those strains.

Aspergillus oryzae has a strong phosphatase activity. FAD is hydrolyzed first to FMN with the enzyme and then quantitatively to riboflavin and inorganic phosphate. The latter reaction would be carried out with phosphomonoesterase II, this inference being different from the Warburg theory. The properties of the enzyme was investigated employing Takadiastase directly. Reaction velocity is linear to time at least during a certain period from the beginning and Michaelis-Menten constant Km is 1.28×10^<13> mol. Activity of the enzyme shows apparent decrease at pH 3.2 and 6.4. F', Ag' and molybdates inhibit the enzyme action and Mg" has neither inhibiting nor activating effect. The inhibition observed with Ag ion seems to due to the formation of riboflavin-AgNO_3 complex. Competitive inhibition with β-glycerophosphate was not observed in the concentration less than 10^<-2> moles per litre. Riboflavin-AgNO_3 complex has an absorption maximum at 267 mμ and riboflavin at 265 mμ.

The author investigated the influence of varying the components of Pfeffer solution on the production of flavinmononucleotide (FMN) and flavinadeninedinucleotide (FAD) by a strain of Aspergillus oryzae. FAD in mycelium greatly decreased and FMN gradually decreased with advancing days. The addition of riboflavin to the media seemed to cause no difference in the amounts of FMN and FAD. The presence of Fe‥, seemed to decrease the amounts of the both substances with the increasing concentrations. In case of Fe…, the optimum concentration was found to be ca. 10^<-5> moles per litre. The carbon source whatever kind it may be, exerted little influence, but 1 per cent of alcohol produced relatively larger amounts of FMN and o-ribose always gave low valus. As a nitrogen source, NO_3 was much superior to NH_4. No goood results were obained with organic nitrogen sources.

Investigations were made on the excretion of pyridoxine plus pyridoxamine, pyridoxal and 4-pyridoxic acid in the urine of patients with various diseases as well as healthy infants, children, and adults during 24 hour with and without the administration of pyridoxine. The results showed that a considerable diferrence was recognized in the urinary excretion of B_6 vitamins and their metabolite among individuals and days as well as a seasonal fructuation, showing a tendency to decrease in hot season. The maximum excretion of pyridoxine plus pyridoxamine in urine was recognized 1 hour after the administration of pyridoxine, and those of pyridoxal and pyridoxic acid followed the former, and their excretion returned to normal in the same order. Abnormal vitamin B_6 metabolism was observed in the children who suffered from systemic eczema, asthma, apparent ariboflavinosis, acute nephritis or nephrosis.
